چکیده

Variation of Alternaria brassicicola leaf spot of Broccoli (Brassica oleracea var. italica L.) was evaluated based on morphological, cultural and molecular parameters during Rabi season 2010 and 2011. Six isolates of Alternaria were cultured in vitro using different growth medium and the growth pattern of the fungi was studied. Maximum 86 leaf spots were recorded from Naini followed by BHU (74) and minimum 17 spots from CSA samples. The size of spots shown variations in different isolates collected from different places. The leaf spots were followed the reducing trend with the increasing number of spots. The maximum 0.5-1.9mm size of spot was noticed in CSA samples and minimum 0.2-0.7mm from Naini sample. The colour of spots was light olivacious, brown with concentric rings with black pin point at the centre. The conidial length ranged 13-120 μm, width 6-16 μm with 2-8 septa. The growth of A. brassicicola recorded maximum in host extract media followed by Potato Dextrose Agar while the growth observed minimum in Czapek’s medium. The variations also exhibited in protein profiling by using SDS-PAGE. The highest molecular weight of protein was detected in CSA (22kDa) isolates followed by isolates from ND (21kDa) and BHU (21kDa) while minimum 20kDa of protein was recorded in Naini isolate.

Shoot-root traits of broccoli (Brassica oleracea var. italica L.) as influenced by different irrigation schedules

Frequency of irrigation to the crops is the key factor for proper growth and economic yield. Therefore, in the current study growth of broccoli plants was investigated under different irrigation levels in pot culture condition. Five levels of irrigation schedules were applied including I0 [Non-irrigation control], I1 [Irrigation up to 15 days after transplanting (DAT)], I2 [Irrigation up to 30 ...

اثر بستر‌های مختلف کشت و شوری بر رشد و عملکرد کلم بروکلی (Brassica oleracea var. italica) در سیستم کشت بدون خاک

Salinity is a very serious problem for agricultural development, especially in arid and semi-arid regions. On the other hand, many benefits of soilless culture have caused the expansion of these systems in these areas. In this research, in order to investigate the effects of different substrates and salinity stress on physiological characteristics and yield of broccoli, an experiment was conduc...

Compositional and Proteomic Analyses of Genetically Modified Broccoli (Brassica oleracea var. italica) Harboring an Agrobacterial Gene

Previously, we showed improved shelf life for agrobacterial isopentenyltransferase (ipt) transgenic broccoli (Brassica oleracea var. italica), with yield comparable to commercial varieties, because of the protection mechanism offered by molecular chaperones and stress-related proteins. Here, we used proximate analysis to examine macronutrients, chemical and mineral constituents as well as anti-...

Chromosome Doubling of Microspore-Derived Plants from Cabbage (Brassica oleracea var. capitata L.) and Broccoli (Brassica oleracea var. italica L.)

Chromosome doubling of microspore-derived plants is an important factor in the practical application of microspore culture technology because breeding programs require a large number of genetically stable, homozygous doubled haploid plants with a high level of fertility. In the present paper, 29 populations of microspore-derived plantlets from cabbage (Brassica oleracea var. capitata) and brocc...
